You cannot do it server-side. Use the auto-reply functionality or configure an Outlook rule.

You can use transport rules several ways. look into below examples

Hi,

What about creating a transport rule to block messages sent to example.com with explanation?

Go to EAC > mail flow > rule. Choose More Options, and create a new rule like: "If the sender is outside the organization, and the recipient address includes 'contoso.com', reject the message with the explanation ..."

Here is the test result:

Please pay attention that, "The recipient address includes" condition only matches messages that are sent to the recipient's primary email address.

Hi,

Based on my knowledge, we can not only use transport rules to reach this requirement and need to use Inbox rule.

We can create a transport rule like this: "When the sender is outside and recipient address includes 'contoso.com', prepend the subject with 'External to contoso.com' "

Then create an inbox rule like: "When receive a message with 'External to contoso.com' in the subject, reply with a template".
