We have DFSR setup between two 2012 Servers, for DR purposes only. We do not have a Namespace setup yet.
Replication Group is configured and replication is working correctly.
Can we change the Share Name of one of the folders without affecting replication?
If not, what do we need to do to change the share name and have replication work correctly?

You cannot rename a shared folder in DFS namespace. You will get an error regarding removing the folder from namespace first.
So you should delete the folder target(s) in DFS namespace, rename the namespace, rename folders and their shared names, add them back to DFS namespace.
As I said above, we have no Namespace setup yet.

I don't need to change the folder name, just the share name.

You mean the share name under the dfs namespace, not the folder name under the specific drive?
If so, that shouldn't broke your current replication.

Again, no Namespace has been setup yet. The Share is replicated via DFS Replication Group.

If so, as Nedim said, you could consider to rename the file folders, recreate the server shares, retarget the DFS share.
