I've received the following update error below:
Update for Windows Defender - KB2267602 (Definition 1.237.316.0) - Error 0x80070643
How to fix this issue?

I updated Windows Defender and then tried again. Worked for me.

Do you have any issue with system restorem point, cuz i got - Details:
System Restore failed while restoring the directory from the restore point.
Source: AppxStaging
Destination: %ProgramFiles%\WindowsApps

I found another duplicated case of yours about this issue, one of our colleagues Teemo has replied there. We have submitted it via our internal feedback tool on our side. You could use the Windows 10 built-in "Feedback" tool to submit the issue on your side. I hope it could be fixed in the near future.

And me too, same error.
And when updating from Win Defender itself it complains that it 'The virus and spyware definitions didn't update on 28-Feb-2017, 09:11 because of an Internet or network connectivity problem.'

Nothing, Microsoft solved that problem. Try update definition.

Give this a try.................
Open Elevated Command Prompt > Copy/paste this command line..........
"%PROGRAMFILES%\Windows Defender\MPCMDRUN.exe" -RemoveDefinitions -All
> Enter.
Then, in the same Elevated Command Prompt, copy/paste this command line...........
"%PROGRAMFILES%\Windows Defender\MPCMDRUN.exe" -SignatureUpdate
> Enter.
Now try updating your Windows Defender again.

Tried what you are suggesting (david hk129). Still no luck. Command line tool gives the same error.
I am getting this error on my eight server 2016 installations.
Think MS need to get in to this.

same... updater install the 1.237.77.0 but not the 1.237.316.0 update.
