I have created a new web application in sharepoint 2010 , when i browse the created web application i am receiving the below error .

This error (HTTP 404 Not Found) means that Internet Explorer was able to connect to the website, but the page you wanted was not found. It's possible that the webpage is temporarily unavailable. Alternatively, the website might have changed or removed the webpage.

I have tried deleting the web applicaiton and granting permissions to all accounts which are related and also checked the iis related application pool , running users permissions for the related services tooo .

when i create top level site collection the site collection is working fine But the home site is not working 

Windows 2008 R2

Sharepoint 2010 

Sql Server 2008 R2

like 

Ex: http://xxxxxxxxx/    - Is not working 

 http://xxxxxxxxx/sites/employeeservices/ is working fine 

Please help me if any body has solution for it .

You need to create the site collection at the web application root site in addition to the site collection you already created. When you create the site collection, just make sure you don't use a manage path for the web application and your site collection will be created at the root.
