I need to be able to stop notifications from displaying while staff are duplicating their screen and presenting a PowerPoint.
Firstly, will changing the option in Focus Assist 'When I'm duplicating my Display' to ON stop pop ups while presenting does this also work if they are in Extend mode?
Secondly, how to stop Focus Assist notifications themselves from popping up?
Thirdly, how to do this via a GPO or Registry?

First question: I think Yes.
Second question: go to Settings > System > Focus Assist, do what you want to do.
Third question:
Navigate to User Configuration > Policies > Administrative Templates > Start Menu and Taskbar > Notifications.
The following polices may be of interest to you:
Set the time Quiet Hours begins each day -- Start time for the automatic rule.
Set the time Quiet Hours ends each day -- End time for the automaticrule.
Turn off Quiet Hours -- Disables the feature.
Turn off calls during Quiet Hours -- Calls will be blocked automatically if Focus Assist is enabled.
Microsoft renamed and redesigned the Quiet Hours feature of the Windows 10 operating system with the release of the April 2018 Update for the operating system. This is why you will see the words "Quiet Hours" rather than "Focus Assist" when configuring your GPO.
